Studies on Fine Structure and Enzyme Activity of Toxoplasma gondii in Macrophage

Abstract
Micromorphology and enzyme activities of Toxoplasma gondii in macrophage were studied.
One ml of sterile liquid paraffin was injected into peritoneal cavity of mouse five days prior to Toxoplasma inoculation to stimulate macrophage production in peritoneal fluid. Two or 3 days ,after the Toxoplasma inoculation, the peritoneal exudates were aspirated, and the isolated Toxoplasma or macrophages which contained parasites were subjected for micro-morphological and enzymatic studies.
The infra- and extracellular trophozoites of T. gondii had pellicle with an outer and inner membrane, a conoid, micronemes, elongated rhoptries with bulbous alveolar posterior ends, a nucleus, osmiophilic bodies and lipoid bodies. The lipoid bodies scattered in cytoplasm of Toxoplasma, but tended to get together after engulfed in macrophage. A parasitophorous vacuole developed in later stages of the parasite, and membranous folds and stereocilia were observed in it. Stereocila in the vacuole and the thread-like filaments seemed to be originated from Toxoplasma. Ribosome-like granular materials in macrophage gathered into the parasitophorous vacuole, and mitochondria of the host cell lined up around the periphery of the vacuole.
Acid phosphatase activity was demonstrated in the two-layered anterior part of the pellicle, contents of the parasitophorous vacuole and phagosome-like vacuoles. Peroxidase activity was. demonstrated in the outer membrane of rhoptries of the parasite and the cristae of the mitochondria which lined close to the parasitophorous vacuole.
