Abstract
This study was performed to observe the ultrastructure of Acanthamoeba culbertsoni and Naegleria¢¥ fowled with the special reference to the enzyme activity. The amoebae were grown axenically at 37¡ÆC in the CGVS medium (Willigert, 1971). For demonstration of acid phosphatase Gomori medium (Eranko et al., 1952) was used, and method by Cotran and Litt (1970) for peroxidase activity.
Numerous vacuoles of three different structure in size and shape were observed to be contain remnant of degenerated food particles in the Acanthamoeba culbertsoni. The rough endoplasmic reticulum, some of ivhich was associated with small vacuoles, was shown in the trophozite of Naegleria fowleri. Mitochondria comprising typical cristae were abundantly distributed all over the .cytoplasm in Acanthamoeba culbertsoni and also in Naegleria fowleri.
Reaction product of acid phosphatase was present in aggregates in the membrane of .and within vacuoles, especially in the large vacuoles. But peroxidase reaction product was not observed in the trophozoites of Acanthamoeba culbertsoni and Naegleria fowleri.
