Synthesis and Hyperglycemic Actibity of N^(G)-Monoalkylagmatine Derivatives

Abstract

N^(G)-Monoalkylagmatine derivatives, the decarboxylation products of N^(G)-monoalkyl-L-arginine dervatives, have been synthesized by coupling putrescine and N-alkyl-S-methylthiopseudouronium iodides which were produced from monoalkylthiourea and methyl iodide.
The monoalkylthioureas which are not available commercially were obtained by reacting monoalkylamines, carbon disulfide, ammonia, and ethyl choroformate with good yields.
The agmatine derivatives were purified by ion exchange resin column chromatography with a gradient of 1~5 HCl and recrystallizing their picrates from water.
The blood glucose levels in rats affer intraperitoneal injection of N^(G)-monoalkylagmatines increase in order of N^(G)-ethyl-, N^(G)-propyl-, N^(G)-butyl, and N^(G)-pentylagmatine.
