Comparison of Action of Cocaine and Desmethylimipramine

Abstract
1. The influences of cocaine and desmethylimipramine (DMI) were investigated in the cardioaccelerating action of norepinephrine, epinephrine, isoproterenol, tyramine, DMPP and McN-A-343 in spinal rabbits, the positive inotropic action of norepinephrine, tyramine, DMPP in the isolated muscle preparations of rabbit atria and ventricles, the pressor action of norepinephrine, epinehprine, tyramine and DMPP.
2. Both cocaine and DMI potentiated the cardioaccelerating action of norepinephrine.
3. Cocaine did not affect the cardioaccelerating action of epinephrine, isoproterenol, tyramine, DMPP and McN-A-343. DMI potentiated the accelerating action of epinephrine but weakened the accelerating action of isoproterenol, tyramine and McN-A-343. It did not affect the action of DMPP.
4. Cocaine, but not DMI, potentiated the positive inotropic action of norepinephrine in both the atrial and ventricular muscles. Cocaine and DMI weakened the inotropic action of tyramine and, DMPP in both muscle prepations.
5. Cocaine and DMI potentiated the pessor action of norepinephrine, epinephrine and DMPP but weakened that of tyramine.
6. The above results suggest the difference in action manner of cocaine and DMI in the sinoatrial node, cardiac muscle and vascular muscle.
