Effect of Agaricus biorenovate Extract on Collagen Synthesis and Matrix Metalloproteinase-1 Production in Human Dermal Fibroblast

Agaricus blazei, a mushroom, has been traditionally used not only as a medicine to treat diseases such as cancer, diabetes, and high blood pressure, but also as a food. However, the anti-wrinkle and anti-aging properties of Agaricus blazei extract has not been elucidated. Here we evaluated the effect of Agaricus blazei extract and its biorenovation product on the production of metalloproteinsase-1 (MMP-1) and type 1 collagen in TNF-¥á-treated human dermal fibroblast (HDFs). We identified the newly emerged peaks of biorenovation product on HPLC chromatogram. Relative to its nonbiorenovation control group, the proliferation and cell viability in biorenovation product-treated HDFs were increased across the concentration range of sample treatments. Our results also showed that biorenovation product significantly suppressed the production level of MMP-1 but conversely increased the secretion level of type 1 collagen as compared with those of controls. In this study, we reported the significant improvement of anti-wrinkle and anti-aging properties of Agaricus blazei extract through biorenovation process, which would in turn facilitate the research and development process of cosmetic products.
