Anti-obesity effect of black ginger (Kaempferia parviflora) extract in 3T3-L1 adipocytes and high fat diet-induced obese C57BL/6J mice

Abstract

Black ginger (Kaempferia parviflora Wall. Ex Baker), belonging to the family Zingiberaceae, has long been employed as a functional food ingredient due to its antioxidant and anti-inflammatory activities. This study investigated the anti-obesity effect of K. parviflora ethanol extract (KPE) in differentiated 3T3-L1 cells and high-fat-diet (HFD)-induced C57BL/6J mice. Administration of KPE 100 or 200 mg/kg/day reduced body weight, adipose tissue weight, adipocyte size, and fat volume of HFD-induced obese mice. At the molecular level, KPE downregulated the adipogenic transcription factors, including peroxisome proliferator-activated receptor ¥ã (PPAR¥ã), CCAAT/enhancer binding protein ¥á (C/EBP¥á), sterol regulatory element-binding protein-1c (SREBP-1c), and lipid synthesis enzymes, such as fatty acid synthase (FAS) and lipoprotein lipase (LPL) in 3T3-L1 cells and C57BL/6J mice. Additionally, KPE activated AMP-activated protein kinase (AMPK) pathways and fatty acid oxidation. Collectively, these results suggest that KPE could be used as a functional agent for attenuating obesity.
