Association of VEGF gene polymorphism with diabetic retinopathy in type 2 diabetes mellitus

Abstract
Purpose: Vascular endothelial growth factor (VEGF), a major mediator of angiogenesis, may play an important role in the pathogenesis of diabetic retinopathy. The aim of this study was to evaluate the effect of the genetic variations of VEGF gene to assess its possible association to diabetic retinopathy in type 2 diabetic patients. We also examined the effect of genotypic variation of VEGF (C634G) on diabetic nephropathy and macrovascular complications such as coronary heart disease and stroke.

Materials & Method: We conducted a case-control study, which enrolled 228 patients with type 2 diabetes. A total of 166 patients with diabetic retinopathy were compared with 62 patients without diabetic retinopathy. VEGF genotyping was determined using polymerase chain reaction with restriction fragment length polymorphism using BsmF1 restriction enzyme.

Results: Distribution of VEGF (C634G) genotype in the patients either with retinopathy or without retinopathy was confined to Hardy-Weinberg equilibrium. The patients with retinopathy have higher serum BUN and creatinine levels than the patients without retinopathy. There was no evidence of association between VEGF (C634G) genotype and the presence of diabetic retinopathy in type 2 diabetes. Diabetic nephropathy and macrovascular complications such as coronary heart disease or stroke were not associated with C634G polymorphism of VEGF gene either.

Conclusions: These results suggest that C634G polymorphism of VEGF gene was not associated with diabetic retinopathy in type 2 patients. Further studies are required to approve the role of genetic susceptibility in the development of diabetic retinopathy.
