Establishment of Human Papilloma Virus 16 (HPV 16) Positive Mouse Tonsil Cancer Cell Line and Treatment of HPV 16 Positive Mouse Tonsil Cancers with Radiation or Vaccination against E6/E7

Abstract
Purpose: The role of human papilloma virus (HPV) 16 in malignant transformation has predominantly been investigated in anogenital keratinocytes. The author tried to establish HPV 16 positive mouse tonsil cancer cell line and treat the tumors with radiation or vaccination against HPV 16 after implantation of the cancer cells into syngeneic mice.

Materials and Methods: Normal male C57BL/6 mice tonsil epithelium was dissected and mouse tonsil cancer cell line was established by transducing mouse tonsil epithelial cells with HPV 16 E6 or E7. After implantation of mouse tonsil cancer cells into syngeneic mice, the tumors were treated with radiation of 20 Gy at day 31 or vaccination against HPV 16 E6/E7 at day 28.

Results: Mouse tonsil epithelial cells were immortalized when transduced with HPV 16 E6. But, in addition to E6, H-ras was required for tumor formation. Treatment of HPV 16 positive mouse tonsil cancers with radiation, vaccination or vaccination followed by radiation did not show any tumor shrinkage and there was no mouse survived.
Conclusion: Further in vivo experiments including chemotherapy, combined chemoradiation therapy or surgical excision will be needed in order to establish more proper treatment methods for HPV16 positive tumors.
