Gene Therapy; Basic Principles and Clinical Applications

Abstract
Since 1989 when clinical trial of gene therapy using retroviral vector was done, gene therapy has widened its application. The possible role of gene therapy has been examined for cancer, cardiovascular diseases, and autoimmune diseases in addition to genetic disorders. The expression of intact protein in the cells through the transduction of normal gene into the cells is the central issue in gene therapy. In this point, the efficacy of gene transfer and the choice of target cells are very important. We reviewed here the characteristics of gene transfer methods, and discussed the prospect of gene therapy through the review of current clinical trials.
