Identification of the Transcription Regulatory Sequences for the Nitrogen Catabolite Repression of Yeast ASP3 Gene

Abstract
Asparaginase II negative Saccharomyces cerevlSlae strain, XE223-1D, was transformed with plasmids pKW1 (8.2-kb insert), pKW2 (4.2-kb insert) and pKW3 (4.0-kb insert) which contain the ASP3 gene and its flanking sequences. Plasmids pKW1 and pKW3 conferred a normal level of asparaginase II biosynthesis and a normal nitrogen catabolite repression (NCR) to their host strain. In contrast, plasmid pKW2 was partially resistant to NCR. Plasmid pKW2 carries a small deletion (350 bp) in the upstream region of the ASP3 gene. Therefore, we suggest that NCR-regulatory sequences of the ASP3 gene are located in the upstream region of the gene.
