The Structure of Two Additional Human Serine tRNA Genes

Abstract

|
Two human serine tRNA(AGA) genes have been isolated from different recombinant ¥ë phage clones harboring human DNA The nucleotide sequences of the genes and their flanking regions have been determined. The sequence analyses revealed that the tRNA coding sequences of the two genes and more than 100 bp of their 5'-flanking regions are identical, while their 3'-flanking sequences are different, suggesting that the two genes may have arisen by duplication. An Alu sequence was found about 400 bp downstream from one of the tRNA genes. The tRNA genes were transcribed in an in vitro transcription reaction using a HeLa cell nuclear extract, suggesting that the two human serine tRNA genes could potentially be functional in vivo.
