Effects of Estrogen and Dopamine on Prolactin Synthesis in the Rat Pituitary

Abstract

The effects of estrogen;.and dopamine on prolactin synthesis at a single-cell level were examined using immunohistochemistry combined with in situ hybridization histochemistry. Estrogen implants (235 pg/ml) to adult ovariectomized rats significantly increased the number of lactotrophs and the level of cellular prolactin mRNA in the lactotrophs compared with those of the ovariectomized rats. In response to a dopamine receptor agonist bromocriptine and an antagonist haloperidol, number of immunoreactive lactotrophs and prolactin mRNA level were not substantially altered. However, haloperidol significantly increased the serum prolactin level, while bromocriptine suppressed it. These data show that estrogen not only causes proliferation of lactotrophs but also increases prolactin synthesis per cell and that dopamine affects prolactin level through regulation of its synthesis or secretion.
