Molecular cloning and Nucleotide Sequence of cDNA Encoding the Precursor for Soybean Trysin Inhibitor (Kunitz Type)

|
cDNA clones encoding the precursors for the soybean Kunitz trypsin inhibitor (SKTI) were isolated and characterized. A cDNA expression library was constructed with mRNA isolated from developing soybean seeds. The clone M-31, a near full-length cDNA clone for SKTI-a, was isolated by immunological screening with anti-SKTI antibody. Nucleotide sequencing of the clone shows one open reading frame of 651 by which encodes 217 amino acids for the SKTI-a precursor. There are 12 by of Y-noncoding and 113 by of Y-noncoding sequences containing two polyadenylational signal sequences, "AATAAA," and a part of the poly(A) tail. Deduced amino acid sequence of the open reading frame matches to the amino acid sequence of the mature SKTI-a from seeds. The cDNA clone for SKTI-a encodes extra 25 amino acids at the amino terminal and a stretch of 11 amino acids at the carboxy terminal. It suggests that the precursor is processed at both the amino terminal and carboxy terminal by post-translational modification to produce mature SKTI in soybean seeds. Another cDNA clone, XS-4, was isolated by nucleic acid hybridization screening of the cDNA library with the clone M-31 as a probe. The clone 7S-4 encoding SKTI-b shows nearly the same structure as 1,,Ti-31 except 11 nucleotide difference, which results in the 9 amino acid difference from SKTI-a. These genes are expressed only in seeds ¢¥ but not in leaves, maintaining a tissue specific gene expression pattern which is regulated at the level of transcription. Genomic Southern blot analysis demonstrates that these genes are encoded by relatively small number of gene family.
