Sequence analysis of the Promoter Region of the Human ¥è1 Globin gene

Abstract

|
The human 01 globin gene identified recently is located at the very 3¢¥ end of the globin a gene cluster, approximately 3-kb downstream from the adult al globin gene. It is shown here that the upstream promoter region of the human 01 globin gene consists of a novel arrangement of Alu family repeat sequences in which the CCAAT box is contained within a 38-bp internal segment of Alu repeat sequence. Immediately upstream from this CCAAT box containing Alu repeat segment is a 241-bp Alu repeat pointing in the opposite direction. The first promoter element of Alu segment of the 01 gene is matched exactly to the box A of Alu consensus sequence, while the second promoter element has two transition mismatches to the box B of Alu consensus sequence. This is the first example of an Alu repeat sequence to function as the promoter of polymerase II-dependent gene(s), and its possible regulation by another Alu repeat sequence located immediately upstream.
