A Site-specific Deletion Mutant of Murin ¥ã-Interferon Showing Anti0Viral Activity in Vitro
Lee Young-Ik
Abstract
Four amino acids were deleted in murine y-interferon gene using simple oligonucleotide directed mutagenesis in order to check whether the 4 amino acids were required for the anti-viral activity. Plasmid pYL-2 was constructed using expression vector pEV-vrf3 and the plasmid pMifko-1 which contains the whole coding sequence for the cDNA and 5¢¥-and Y-noncoding sequences. Results demonstrated that deletion of the four amino acids from the amino-terminal region of the murine y-interferon still showed anti-viral activity in vitro.
