Caffeine Induces a round Cell Morphology in Early S Phase NIH 3T3 similar to That of Mitotic Cells

Abstract

We have previously shown that overexpressed chicken pp60`-5f has retarded gel mobility and enhanced kinase activity during NIH 3T3 cell mitosis [Chackalaparampil, I., and Shalloway, D. (1988) Cell 52, 801-9021. Here we show that kinase activity of pp60 increases in early S phase cells by caffeine with a morphological change similar to that of mitotic cells. However, there are no mobility retardation on sodium dodecyl sulfate polyacrylamide gel electrophoresis and novel phosphorylation. These results suggest that enhanced kinase activity of pp60`-"` may account for the round cell morphology at least in part.
