Aphidicolin-sensitive DNA Polymerase Plays a Major Role in DNA Repari Synthesis Induced by UV Light in HeLa Cells

Abstract

|
DNA repair synthesis induced by ultraviolet light in HeLa cells in which DNA replicative synthesis had been highly suppressed was inhibited independently by aphidicolin (an inhibitor for DNA polymerases a and S) and 2¢¥,3¢¥-dideoxythymidine (ddThR, an inhibitor for DNA polymerase 0). The incomplete repair patches accumulated during incubation with aphidicolin were reversibly sealed upon the replacement of aphidicolin with ddThR, whereas those accumulated during incubation with ddThR were not sealed upon replacement of ddThR with aphidicolin. These results indicate that aphidicolin-sensitive DNA polymerase (a or S) can complete the repair synthesis without the participation of ddThR-sensitive DNA polymerase 0 and the two polymerases do not act sequentially - on the same repair sites.
