Cytotoxicity of Human tumor Necrosis Factor lacking 14 N-terminal Amino Acids Produced in Escherichia coli

Abstract

Human tumor necrosis factor (TNF) gene was isolated from Xgtll genomic library of human leukocyte and cloned in E. coli using exon fusion method. The recombinant TNF molecule corresponding to the third and fourth exons of human TNF gene was produced in E. coli at high level, and this recombinant protein shows high cytotoxic activity against murine L929 cells. Furthermore, the recombinant TNF, of which 14 amino acids of the N-terminus was substituted by 34 amino acids of P-galactosidase N-terminus, also revealed cytotoxic activity. This result suggests that the N-terminal 14 amino acid sequence is not essential for the cytotoxic activity of human TNF.
