Genomic Cloning of Mouse Cellular retinol binding Protein Gene

Abstract
The gene for the cellular retinol-binding protein (CRBP) was cloned from a mouse genomic library of lambda Fix II. Two overlapping clones which together encompass about 19 kb were isolated from an initial screening of the library. Restriction mapping and Southern analysis using three oligonucleotide probes derived from the cDNA localized the coding sequence within the 9-kb HindIII and the 4.2-kb BamHI fragments. These fragments have been subcloned and sequenced by the modified dideoxy chain termination method using 5-methyl dCTP instead of dCTP. The size of the CRBP gene is about 9.5 kb and it consists of four exons. The second intron occupies 6 kb of the 9.5-kb gene.
