Growth Phase and Tissue Specific Variations in the Multiple Murine Dihydrofolate Reductae mRNAs

|
Mammalian cells contain multiple dihydrofolate reductase (DHFR) mRNA species which are transcribed from the single dhfr gene. Four major mRNA species of 1.6 kb, 1.2 kb. 1.0 kb, and 0.75 kb are found in mouse cells. We have analyzed the relative abundance of the multiple DHFR mRNA species in mouse A9 cells in different growth phases, exponentially growing or stationary phase, and also in various tissues. The 1.6-kb and 0.75-kb mRNAs are almost equally abundant in exponentially growing cells, while the 0.75-kb mRNA is a minor species in stationay cells. The ratio of the 1.6-kb and the 0.75-kb mRNA species varies in different tissues. Our results show that the absolute and the relative levels of the DHFR mRNA species vary in growth- and tissue-specific manners.
