Cloning of Human RNA Polymerae ¥± Elongation Factor TFHS cDNA without the internal Deletion of 21 Amino Acid Sequence

Abstract
We have amplified and cloned the complementary DNA (cDNA) for human RNA polymerase II transcription elongation factor TFIIS using polymerase chain reaction. The size of our cloned human TFIIS cDNA is much larger than that of the clone previously reported [Yoo. O., Yoon, H., Baek, K., Jeon, C., Miyamoto, K, Ueno, A., and Agarwal, K (1991) Nucleic Acids Res. 19, 1073-1079]. The primary structure of human TFIIS was deduced from the nucleotide sequence analysis of our cDNA clones. It contains the 21 amino acid sequence which was previously reported to be deleted in the middle by Yoo et al. (1991). Thus, our data suggest that the human TFIIS protein consists of 301 amino acids like the mouse TFIIS protein. Using the cDNA as a probe, we analyzed the human genomic DNA for TFIIS gene. Southern blot analysis demonstrated that TFIIS is coded by a single copy gene, rather than multiple gene family.
