The Effect of ¥â-Octyl Glucoside on the Crystallization of Hen Egg white Lysozyme

Abstract

|
The effect of O-octyl glucoside on the crystallization of hen egg white lysozyme has been examined in order to investigate the possible structural changes induced by binding of the nonionic detergent to proteins. The most dramatic effect resulting from the addition of P-octyl glucoside was the growth of tetragonal crystals under orthorhombic crystallization condition. X-ray diffraction experiments were performed on tetragonal crystals grown under the orthorhombic crystallization condition in the presence of P-octyl glucoside. The space group is P4,212 (or P43212) with unit cell dimensions similar to those reported for the conventional tetragonal crystals. Unit cell dimensions and the sensitivity to synchrotron X-rays showed a slight variability depending on the concentration of Q-octyl glucoside. X-ray data to 1.9 A Bragg spacing have been collected using synchrotron X-rays.
