Cultured Myoblasts Secrete a Protein That Is Capable of Promoting Myogenic Differentiation

Abstract

|
A protein capable of promoting myoblast fusion was purified to homogeneity from muscle-conditioned culture medium using conventional chomatographic procedures. This fusion promoting factor (FPF) has a native size of 500 kDa and consists of two polypeptides with sizes of 175 and 145 kDa. Both polypeptides have an isoelectric point of 5.5. When the purified FPF was added to culture medium, myoblast fusion initiated appriximately 6 h earlier than normally occurred. In addition, FPF promoted the accumulation of muscle specific proteins, such as acetylcholine receptor. Furthermore, treatment of anti-FPF IgG inhibited both the morphological and biochemical differentiation of cultured myoblasts. These results suggest that FPF acts as a positive regulator for myogenic differentiation of embryonic muscle cells in culture.
