Molecular Cloning of Genomic DNA Encoding the Soybean Glycinin subunit A2B1

Abstract
A nearly full-length cDNA clone and a genomic clone for soybean glycinin genes were isolated to study the regulatory expression mechanism of the plant gene. A soybean genomic DNA library was constructed in EMBL3 vector with soybean total DNA isolated from soybean leaves. Genomic DNA clones for glycinin subunits were isolated by plaque hybridization of the library with the cDNA clone for glycinin subunit A2131a as a probe. A genomic clone XG47 was isolated from the library and restriction enzyme mapping was carried out by Southern blot analysis with cDNA clones, pG1Al-L and pG1A1-S, which encode the C-terminal and N-terminal of the glycinin subunit A2131a, respectively. It demonstrated that the clone X.G47 contains two glycinin genes, Gyl and Gy2, in the same orientation with an intergenic region of 2.7 kb. The structure of the glycinin gene Gy2 was determined by nucleotide sequencing. The Gy2 gene consists of 4 exons and 3 introns, spanning over 2,863 by and encodes 485 amino acids. Nucleotide sequences of the two genes show 85% nucleotide sequence homology at coding region and 95% at 5¢¥ flanking region, -170 to -1. The transcription initiation site of Gy2 gene was determined to be at 43 by upstream of translation initiation site, ATG.
