Effects of Activated c-H-ras Oncogene Expression on the Growth of Androgen-dependent Mouse Mammary Carcinoma SC115 Cells

Abstract
To investigate the role of oncogenes in the progression of hormone-dependent cancers, the activated c-H-ras oncogene was stably transfected into an androgen-dependent cell line, SC115-A7, cloned from a SC115 tumor. A clone acquiring the activated c-H-ras oncogene, SC115-ras4, showed transformed phenotype and increased tumorigenicity, but was still androgen-dependent for maximal growth in vivo and in cell culture. Effect of ras oncogene on the expression of fibronectin and collagen genes was also investigated by Northern blot hybridization. The RNA levels of these genes decreased coordinately after androgen treatment in parental SC115-A7 cells, but increased in SC115-ras4 cells. These results suggest that the ras oncogene may not be a direct cause of the progression in SC115 tumor. However, ras oncogene may alter the regulation of gene expression by androgens; causing a certain kind of indirect effect in the progression.
