Dissolution of Polyhedral inclusion Bodies of hyphantrai cunea Nuclear Polyhedrosis Virus

Abstract
The action of an alkaline protease on the polyhedral inclusion bodies (PIB) of the Hyphantria cunea nuclear polyhedrosis virus (HcNPV) which was replicated in the Trichoplusia ni cell line (TN-368), was studied using the dissolution kinetics of HcNPV with the treatment of an alkaline solution. The degradation of PIB occured faster at the higher pH and the rate of PIB dissolution increased in the presence of NaCl at lower Na2CO3 concentration, while this effect was not significant at higher Na2CO3 concentration. The degradation activity was extensively inhibited by Hg" and Cu-+, but the activity was only 51% inhibited by the treatment of heating at 100 C for 2h. The PIB solution treated with alkali displayed 2-band profile on sodium dodecyl sulfate-polyacrylamide gel electrophoresis and the viral protein showed at least 13 bands ranging from 15.5 to 98.0 kDa.
