Binding of Trans-acting Regulatory Protein DAL82 to Upstream Induction Sequence of Allantoin Degradative Gene DAL7

Abstract
In Saccharomyces cerevisiae, expression of allantoin degradative pathway genes (DAL4, DAL7 and DUR1,2) is induced by allophanate. The regulation of DAL7 gene involves interactions between cis-acting elements, upstream activation sequence (UAS), upstream induction sequence (UIS) and upstream repression sequence (URS), and trans-acting factors coded by the DAL81 and DAL82 genes. In this study, the DAL81 or DAL82 gene was fused with glutathione S-transferase (Sj26) gene and expressed in E. coli. GST-DAL81 and GST-DAL82 proteins produced were tested for their binding activities to the cis-acting DNA elements. Gel retardation assays indicated that DAL81 protein has no DNA-binding activity with any cis-acting elements. Meanwhile, the N-terminal region of DAL82 protein binds to the UIS element. These results suggest that the DAL7 gene expression is induced by binding of DAL82 protein to UIS and also by protein-protein interactions between DAL81 protein and other regulatory proteins.
