Transcription of a DNA Sequence Reducing the Efficiency of the Phage T7 Factor-independent Terminator in Trans

Abstract
Transcription termination efficiency of a phage T7 factor-independent terminator of 41 by has previously been shown by us to vary from 15% to 82%, depending on the transcribed sequences distal to the terminator [Lee, J. T., Kim, H., Moon, K, Kim, S., and Kang, C. (1991) Mot Cells 1, 203-2091 based on the transcription results of various sequence combinations. When two transcription units containing the same terminator were cotranscribed in vitro, either with a two-promoter template or with two one-promoter templates, unexpected variation of the termination efficiency was observed in this study. These results, together with those from transcriptions with various polymerase-to-template ratios, suggest that transcription product of a particular sequence reduce the termination efficiency of the T7 terminator To in trans manner.
