Abstract

|
Chlorophyll a/b-binding (CAB) proteins constitute the light-harvesting complexes in chloroplasts. Their genes are regulated mainly at the transcriptional level by light. A gene encoding CAB protein in potato was isolated and characterized at the molecular level. The cab gene of potato contains an open reading frame coding for 267 amino acids. The amino acids includes 34 amino acids which form the transit peptide involved in transport of the mature CAB protein into chloroplasts. The amino acid sequence of the transit peptide is highly homologous to those of Type II CAB proteins of photosystem II. The 5¢¥ flanking region of the cab gene contains TATATA and CCACT as possible promoter elements. In addition, the region contains the light-responsive elements which have been identified at the region of cab genes of other plants. Genomic hybridization revealed that cab genes in potato are present as a multigene family composed of about 10 members.
