Effects of Phorbol Ester, Forskolin and 8-Bromo-cAMP on Multidurg Resistance Gene Expression

Drug resistance in human cancer is associated with overexpression of the multidrug resistance (MDR) gene, which encodes a drug-efflux pump called P-glycoprotein. Expression of the MDRI gene can occur in human cancers in the absence of drug treatment, but the regulatory mechanism of MDRI gene expression in this phenomenon is not known. In this study, the regulation of MDRI gene expression in human bladder carcinoma cell line J82 was analysed in the presence of tumor promoter or cAMP. We found that 12-0-tetradecanoylphorbol-13-acetate (TPA) can upregulate the MDRI gene expression while cAMP does not have any effect.
