Nucleotide Sequence and Expression of a Chlorophyll a/b Binding Protein Gene in Soybean

Abstract

|
A cDNA clone encoding a soybean chlorophyll a/b-binding protein was isolated and characterized. The soybean cab cDNA clone, designated pGAB3, has a full coding sequence including 31 nucleotides of 5¢¥ untranslated region. Nucleotide sequence analysis of the pGAB3 showed that it contains an open reading frame of 263 amino acids, including a transit peptide of 32 amino acids. The amino acid sequence of the transit peptide and the amino terminal sequence of the pGAB3 indicate that the pGAB3 represents a type I gene encoding a light harvesting chlorophyll a/b-binding protein of photosystem II. Northern blot hybridization showed that the expression of soybean cab genes is induced by light. The expression of soybean cab genes is also tissue-specific; the cab messages are most abundant in leaves and barely detected in roots.
