Isolation and Characterization of Retropseudogene for Human Cytochrome b5

Abstract
From the screening of human genomic libraries, two positive clones were isolated and purified. Southern blot analysis and restriction enzyme mapping showed that they might be related and contain a pseudogene for human cvtochrome bs. From the partial DNA sequencing, this pseudogene showed high homology to human liver bs mRNA, however, it contained many base substitutions and deletions which inactivated itself. Since it did not contain the intronic sequence for the position between amino acids 86 and 87 it turned out to be a processed pseudogene. In addition it was different from the previously reported ones. It is believed to result from the integration of reverse transcripts of cytoplasmic poly(A)+ bs mRNA into human genome by a transposon like mechanism or retroviral activity. The finding of a new processed pseudogene indicates the complexity of genomic organization of human cytochrome bs and will facilitate the design of rapid and accurate diagnostic methods for congenital methemoglobinemia at genetic level.
