Abstract
In Saccharomyces cerevisiae, expression of an allantoin degradative gene, DAL7, is induced by allophanate. The regulation of DAL7 gene expression involves interactions between cis-acting elements including upstream activation sequence (UAS), upstream induction sequence (UIS) and upstream repression sequence (URS), and trans-acting factors such as DAL81, DAL82 and DAL80 proteins. The binding properties of DATA to the cis-acting elements in the DAL7 were investigated by gel retardation and DNase I footprinting experiments using the DAL80 protein which was fused to C-terminal region of glutathione S-transferase (GST). The DAL80 protein fused to GST was demonstrated to bind to UAS and URS2 sequences, which suggested that the DAIM protein may activate transcription of DAL7 both by acting as a GATAA UAS binding factor and by relieving repression. Also, the DAL80 protein seemed to be associated with a 52-kDa protein through protein-protein interaction.
