The relationship between Plasmid-like mtDNAs and Chromosomal mtDNAs in the genus Citrus

Abstract
Mitochondrial DNAs (mtDNAs) from leaves in four species of the genus Citrus (Citrus grandis OSBECK, Citrus 0 TANAKA, Citrus junos SIEB, Citrus unshiu MARC) were isolated and were found to contain three classes of extrachromosomal DNA elements in addition to the chromosomal mtDNA. These plasmid-like mtDNAs which are present in linear double-stranded forms were designated as elements p-0, p-1, and p-2 with approximate sizes of 2.5 kb, 1.4 kb, and 1.0 kb, respectively. To show genetic similarities between plasmid-like mtDNAs and chromosomol mtDNA, we performed DNA-DNA hybridization. As a result, considerable homology was demonstrated between p-1 and other two elements, p-0 and p-2. In addition, homologous sequence appeared to be present between p-1 and chromosomal mtDNA. This suggests that in Citrus plasmid-like mtDNAs p-0, p-1, and p-2 share homologous DNA sequences with one another and also with chromosomal mtDNAs.
