Identification of three Phosphoinositide Specific Phospholipase C from Cytosolic Fraction of Soybean (Glycine max L.) Sprouts

Three soluble enzyme fractions which hydrolyze phosphoinositides were separated from soybean sprouts using matrex green gel and HPLC gel filtration column chromatography. Properties of the three enzyme fractions were different for the requirement of Call and sodium deoxycholate. All were specific to PI, and no enzyme activity was detected for PC or PE under the same assay conditions. Since they produced inositoi phosphate as the major product using [3H] PI as a substrate, it was proved that PLD enzyme was not contaminated to these enzyme fractions. For the full enzymatic activities, the first two PLC fractions required 100 gM Call and 0.05% sodium deoxycholate, whereas the third fraction requested 10 gM Call and 0.025% sodium deoxycholate.
