Effect of Ceramide on c-myc Gene Expression during Diferentiation of U-937 Cells

Abstract
Effects of ceramide on cell differentiation and c myc gene expression were studied in U-937 cells. Treatment of U-937 cells with a cell-permeable ceramide caused a decrease in c-myc mRNA levels in a time- and dose-dependent manner. The level of c-myc mRNA was reduced to 501/o of control in 30 mins by the 6 jiM ceramide treatment and the maximal inhibition of c-myc transcription was observed in 1 h after the addition of ceramidet to 295/o of control. Ceramide treatment also resulted in cell differentiation in U-937 cells when non-specific esterase activites was monitored as a measure of cell differentiation. These results suggest that ceramide plays an important role in the control of cell differentiation.
