Modification of the EBV-based System to Support Episomal eplication in Human Bladder Carcinoma Cell Line EJ

Abstract
Vectors carving the origin of replication of Epstein-Barr virus (EBV) and EBV nuclear antigen I (EBNA-1) have been reported to be maintained extrachromosomally in human cells. However, when we introduced the EBV-based vector p291 containing both the origin of replication of EBV (ori-P) and EBNA-I in the human cell line EJ, it was found to be integrated extensively. In an attempt to devise a way to make it stay episomally in EJ cells, EBNA-1 was separately introduced into the cells prior to the EBV-based vector containing EBV origin of replication. We found that the EBV-based vector p291 was maintained episomally in EJ cells expressing EBNA-I. It appeared that certain level of EBNA-I expression was required for efficient extra-chromosomal replication of EBV origin containing vectors in EJ cells.
