Isolation of a genomic Clone for Cytochrome P450 Oxidase from mentha piperita

Abstract

To study the role of cytochrome P450 oxidase (P450) in menthol biosynthesis of M. piperita, isolation of a gene for P450 was attempted. PCR amplification of genomic DNA of Mentha piperita with two specific primers from conserved regions of P450 gene of avocado fruit gave 423 by long IJNA fragment. Direct molecular cloning and nucleotide sequence analysis of the clone, p2H4, showed an open reading frame encoding 141 amino acids. Nucleotide sequence and deduced amino acid sequence show 61.6% and 58.7% of positional identities to those of P450 cDNA from avocado, respectively. Northern blot analysis shows mRNA size for P450 of about 1,800 nucleotides. Southern blot analysis suggests that P450 gene of M. piperita is organized as a multi-gene family. PCR amplification and Southern blot analysis of genomic DNAs from various plants show the presence of similar genes in other plant species.
