Light Modulated Expression of thioredoxins in Pea Seedlings

Abstract

|
Thioredoxins activate a variety of photosynthetic enzymes including fructose- 1,6-bisphosphatase (thioredoxin f) and malate dehydrogenase (thioredoxin m) in higher plants. Enzyme activities of fructose-1,6-bisphosphatase and malate dehydrogenase were much greater in the crude ex-tracts from light-grown pea leaves than those from dark-grown pea seedlings, suggesting that the activities of both enzymes and/or thioredoxins are modulated by light. light dependent expression of pea thioredoxin f was tested by enzyme activity assay and northern blot analysis. Enzyme activity of the purified fructose-1,6-bisphosphatase from pea leaves was greatly increased by the crude thioredoxin extracts of light grown peas, whereas enzyme activity was unchanged upon addition of the dark grown pea thioredoxin extracts. These results indicate that expression of thioredoxin f might be stimulated by light. Results from dot and northern hybridization as probed by E. coli thioredoxin gene Qm4) show that the expression of pea thioredoxins are controlled by light at the level of transcription.
