Transcription Initiation Sites of Tomato Phenylalanine Ammonia-lyase 5 Gene and Their Usage

Abstract
We have determined the nucleotide sequence and the transcription initation sites of tomato phenylalanine ammonia-lyase: pal (PAL) 5 gene by SI nuclease protection studies. Interestingly, two transcriptioq initiation sites were identified, each corresponding to a different putative "TATA" sequence element with corresponding conserved upstream motifs. The first initiation site was 44 bps and the second was 146 bps upstream of the translation start codon ATG sequence, respectively. When the adult tomato plant was exposed to fight, the shorter transcript, whose basal levels were very low, was preferentially and strongly stimulated. The basal level of longer transcript, however, was quite high and little stimulated by fight illumination. The results strongly suggest that multiple regulatory sites in the pa15 gene promoter may play an important role in response to environmental stress.
