Isolation of a Partial cDNA Clone for Garlic Latent Virus

Abstract
In the previous experiment, we have isolated several cDNA clones for garlic viruses, single stranded linear RNA viruses. We also have purified garlic latent virus (GLV) by serial infection in local lesion host, Vicia faba, and amplified in systemic host, Allium porrum (Choi et al., 1992). To identify the cDNA clone for the GLV, Northern blot analysis of GLV genome was carried out with cl)NA clones for garlic viruses as probes. The result showed that the clone 28-9 is a partial cDNA clone for GLN¢¥. Nucleotide sequening of overlapping partial cDNA clones revealed an open reading frame over the entire sequence of 2,145 by which encodes 715 amino acids. Northern hybridization with either of end-labelled complementary oligonucleotides taken from the middle of the clone 28-9 reveals that the GLV genome is positive stranded RNA. Northern blot analysis with the clone 28-9 demonstrated that the genome of GLV is 8,2 kb long and had poly(A) tail. Nucleotide sequence similarity was observed 55.4% to 718 by of potato virus X (PVX), and 54.5% to 725 by of narcissus mosaic virus (NMV), which encode part of the replicase. Positional identities of deduced amino acid sequence were 61% to PVX and 59% to NMV at the region. It also contains GxxGGKS motif which is conserved in various ATP- and GTP-binding proteins including replicase of PVX and NMV. These results suggest that the clone 28-9 is a partial cDNA clone for replicase of GLV.
