A Dictyostelium discoideum Prestalk Cell-specific Gene in Tansciptionally Repressed by Cyclic AMP

Abstract
The expression of DictyosteGum gene, pDd26 was examined during in vitro monolayer differentiation. The pDd26 gene encodes an mRNA which is highly enriched in prestalk over prepore cells in the migratory pseudoplasmodium. The expression of pDd26 required the stalk cell Differentiation Inducing Factor (DIF) and was inhibited by the presence of caffeine (which inhibits intracellular cyclic AMP accumulation by blocking adenylate eyclase). By in vitro nuclear run-on transcription analysis it was shown that cyclic AMP acts at the transcriptional level to repress the accumulation of pDd26 mRNA.
