Auxin-regulaed and Wound-inducible Expression of a Potato Proteinase Inhibitor I-CAT Gene in Transgenic Tobacco Plants

Abstract

|
The 5¢¥ flanking region of a potato proteinase inhibitor I gene was characterized at the nucleotide level. Furthermore, the mode of expression of the inhibitor I gene was analyzed in transgenic tobacco plants after construction of a chimeric gene between a 2.0 kb fragment containing the 5¢¥ flanking region of the inhibitor I gene and the chloramphenicol acetyltransferase (CAT) gene. The inhibitor I-CAT gene was not expressed in transformed calluses grown on the medium containing auxin, whereas its activity was induced during incubation in the absence of auxin. It was induced in leaves of transgenic plants by mechanical wounding, Wounding of a leaf, moreover, led to systemic induction of the gene in unwounded leaves of the same plant. The wound-induced response was also enhanced by sucrose. These indicated that the potato inhibitor I gene exhibits auxin-regulated and wound-inducible expression and that potato inhibitor I genes are regulated by various regulatory signals similar to potato inhibitor H genes:
