Modulation of detoxifying Potential and Differential Expression of Glutathione Transferase Isoenzyme in the Bacterial ada Gene-transfected Murine Hepatoma Cell

Abstract
In murine hepatoma cells, the expression of glutathione transferase (GST) isoenzyme genes is distinctly shifted from basic type of GST to its acidic form (GST-P). The shift of GST isoenzyme with cellular transformation may implicate the differential survival capacity between normal and transformed cells. In this report, we have tried to compare GST isoenzyme expression pattern between rat hepatoma cells (14-170213) and FTOPKA cells infected with retrovirus carrying bacterial ada gene, encoding O¢¥mG-DNA-methyl transferase, in order to elucidate the biochemical adaptive mechanism of hepatoma cells to anti-cancer drugs and toxic chemicals. FTOPKA cells showed an increased resistance to N-methyl-N¢¥-nitro-N-nitrosoguanidine (MNNG) and mitomycin C than Fr02B cell but not to ethaerynic acid. GST-P isoenzyme was induced in both cells after treatment with TPA. In contrast, the pattern of GST isoenzyme was modified in the FTOPKA cells: that is, increase in the expression of basic GST¢¥s and decrease in that of GST-P. These data suggest that the DNA alkyl transferase gene transfer changes the detoxification potential of rat hepatoma cells and the expression of GST-isoenzyme genes and may affect the tumorigenic property of hepatoma cells in consequence.
