Molecular Cloning of Class I Patatin cDNA

Abstract
A full-length cDNA clone encoding patatin, the major storage protein of potato, was isolated from potato tubers (Solarium tuberosum cv. Sumi) cultivated in Korea. A cDNA library from poly(A)+ mRNA of developing potato tubers was constructed in X011 vector and a full-length class I cDNA clone (pMYQ7) was isolated by plaque hybridization of the library using a class II cDNA from Solarium tuberosum cv. Desiree and class I specific oligonucleotide as probes. Nucleotide sequence analysis showed that it contains an open reading frame of 386 amino acids including a signal peptide of 23 amino acids and there is 93% homology between the cDNA clone isolated and a class I cDNA clone from S. tuberosum cv. Superior (pGM01). Deduced amino acid sequence from an open reading frame of the cDNA clone showed that molecular weight of the protein is approximately 40 kDa and there is 95% homology between the pMYQ7 and pGM01. Northern blot hybridization using pMYQ7 as a probe showed that the expression of the patatin genes is tuber-specific.
