Genetic Variability in the Envelop region of Hepatitis C Virus Siolated from Korean Patients with Non-A, Non-B Hepatitis

The nucleotide sequences of the envelope region of hepatitis C virus containing hypervariable region I and 2 were determined using seven independent cDNA clones isolated from plasma specimens of Korean patients with chronic hepatitis and their amino acid sequences derived from the nucleotide sequences were analyzed. Extensive genetic variations within the envelope region were clustered and interspersed with highly conserved nucleotide sequences. Seven Korean HCV clones could be divided into two groups, typeIl and III, based on the similarities of nucleotide and amino acid sequences. Comparison of these nucleotide and amino acid sequences with those of the American isolate, HCV-1, and the Japanese isolates, HCV-J and HG 36, showed a significant genetic variability: 13.8 to 38.4% and 15.6 to 42.0% at the nucleic acid and amino acid levels, respectively. Sequence conservation specific to subtype was observed in the HVR 2, but not in the HVR 1, of Korean isolates. A portion of E2 was highly expressed up to 10% of the total protein in Escherichia coli. The antigenicity of the E2 region was demonstrated by Western blotting analysis with the serum from a non-A, non-13 chronic hepatitis patient.
