Facile Transformation of Root Explants of Arabidopsis thaliana L. Heynh with a Direct Shooting Medium

Abstract
We describe here a facile transformation procedure of root explants of Arabidopsis thaliana (L.) Heynh. The important features of our procedure are; 1) introduction of a direct shooting media of a single phytohormone combination of 2 mg/1 benzylaminopurine (BAP) and 0.2 mg/I of indole acetic acid (IAA) for conditioning of root explants, for cocultivation with Agrobacteria and for shoot regeneration, and 2) liquid culture of plants in Murashige and Skoog¢¥s medium supplemented with 1% sucrose for easier collection of root material, and 3) use of 2 week-old root material. When applied to two Arabidopsis ecotypes La-0 and WS-0, our procedure generated transformed shoots within 2 weeks after cocultivation (or 5 weeks from sowing) with a transformation efficiency of 33%.
