A Novel Phosphoinositide-specific Phospholipase C in the Cytosolic Fraction of Mature Soybean(Glycine max L.)

Abstract
A novel phospholipase C (PLC) was identified from the cytosolic fraction of mature soybean plant that contains three PI- specific PLC enzymes. In contrast to other PLC isozymes in eucaryotic cells, only one PLC in cytosolic fraction of mature soybean showed a maximum PI hydrolyzing activity in the absence of detergent. Furthermore, its activity was significantly diminished by the addition of sodium deoxycholate (SDC) to the reaction mixture. Half of its activity was reduced by 0.05% SDC. Among the three cytosolic PLCs in mature soybean plant, the binding force of this enzyme to Matrex green gel was stronger than the others. Therefore, it was eluted last from the gel by 430 mM KCI. Since it was expressed not in soybean sprouts grown in the dark, but in mature soybean plant cultured in the light, it might be suggested that fight is necessiated to express this enzyme. Its activity showed Cat+ dependency and maximum activity was attained at 1 mM free CaZ+ without the addition of SDC to the reaction mixture. In contrast, the other two PLCs in mature soybean plant had no activity without SDC, but they showed maximum PI-hydrolyzing activity at 100 M CaZ+ in the presence of 0.051/o SDC in the assay mixture.
