Heat Shock Elements in Transcription of Mouse HSP70.1 and HSP70.2 Gene

Abstract
The roles of HSE and SV40 enhancer in the promoters of two members of mouse HSP70 family were tested using CAT assay. The promoter of HSP70.2, a member of heat shock
cognate protein, was not heat inducible in mouse fibroblasts. However, it became inducible approximately two-fold by heat shock when the SV40 enhancer was present in the construct. These results suggest that the SV40 enhancer activates single HSE of HSP70.2. Four HSEs of HSP70.1 also enhanced the transcription of HSP70.2 promoter to heat shock. The promoter of HSP70.1, a heat inducible member, showed much higher transcriptional activity than the HSP70.2 promoter even at normal conditions, and induced four-fold of transcriptional level after heat shock. The SV40 enhancer increased transcription of HSP70.1 regardless of heatshock.
