Antioxidant Properties of Protector Protein from Candida against Oxidative Damage of Protein and DNA caused Metal-catalyzed Oxidation System

Abstract

|
An antioxidant protein, the protector, from Candida pseudotropicv& prevented the inactivation of glutamine synthetase caused by the mercaptan/Fe" metal-catalyzed oxodation (MCO) system. In addition the protector inhibited the incorporation of carbonyl groups into protein, fmgmentation of protein, and strand break in DNA induced by the mercaptan MCO system. Those oxidative damages of protein and DNA caused by the ascorbate/Fe" MCO system were not prevented by the protector. In the presence of reduced mercaptan, however, ascorbate/Fell MCO mediated oxidative damages were completely inhibited by the protector. From these results, it is suggested that the protector requires reduced mercaptan as a reducing equivalent to inhibit MCO systems.
